MRI Simulation-guided Boost in Short-course Preoperative Radiotherapy for Unresectable Rectal Cancer
Is NCT03714490 recruiting? Yes, currently enrolling (May 2026). This Phase 2 trial studies CAPOX for rectal neoplasms.
Treatment: CAPOX — Improvements in downstaging are required when using preoperative chemoradiation for unresectable rectal cancer. There is therefore a need to explore more effective schedules. The study arm will receive MRI simulation-guided boost in short-course preoperative radiotherapy followed by consolidation chemotherapy , which may enhance the shrinkage of tumor comparing with the concurrent chemoradiation.

Performance status
ECOG 0–1(Restricted strenuous activity)
Prior therapy
Cannot have received: chemotherapy
No excision of tumor, chemotherapy or other anti-tumor treatment after the diagnosis
Cannot have received: other anti-tumor treatment
No excision of tumor, chemotherapy or other anti-tumor treatment after the diagnosis
Cannot have received: surgery
No excision of tumor, chemotherapy or other anti-tumor treatment after the diagnosis
Lab requirements
Blood counts
White blood cell count ≥3.5 x 10^9/L; Haemoglobin ≥100g/L; Platelet count ≥100 x 10^9/L
Kidney function
Creatinine ≤1.0× UNL; Urea nitrogen ≤1.0× UNL
Liver function
ALT ≤1.5× UNL; AST ≤1.5× UNL; ALP ≤1.5× UNL; TBIL ≤1.5× UNL
